Abstract

Grave-to-cradle: human embryonic lineage tracing from post-mortem body
How a person is made and how each organ develops have been a question from a very long time ago. Though the inherent advantage of in vitro fertilization (IVF) has made it possible for scientists to observe fertilized eggs externally, the 14-day rule, to study embryos through IVF only up to 14 days after fertilization or until the time when a primitive streak occurs, then should be destroyed, was established in 1979. Thus, this 14-day rule has forced human embryo research having no choice but to seek another direction if developmental biologists want to study more than 2 weeks after fertilization, including organogenesis. Here, we discuss the process of conducting retrospective lineage tracing of the human embryology research using the recently developed next-generation sequencing technology especially with a focus on post-mortem human tissues based on physiologic somatic variants. In addition, we highlight the biological and scientific backgrounds of how this was possible, as well as the challenges and hypotheses that need to be solved in the future of human embryology.
